Summary

Aortic stenosis is a common heart valve disease for which no effective drug therapy currently exists. This project aims to better understand the genetic factors that contribute to its development by comparing the genetic profiles of individuals with and without aortic stenosis, and with or without coronary artery disease. The research has identified more than 260 genetic loci associated with the risk of aortic stenosis, uncovered new biological pathways involved in valve calcification, and developed an improved polygenic risk score for disease prediction. These findings may help identify individuals at higher risk and support the development of new preventive and therapeutic approaches.
